Anti-TGF-β1 antibodies are monoclonal antibodies designed to selectively bind and neutralize the TGF-β1 isoform of the transforming growth factor beta (TGF-β) superfamily. TGF-β1 is a pleiotropic cytokine that plays a critical role in immune suppression, tissue fibrosis, and epithelial-mesenchymal transition (EMT). In the tumor microenvironment, TGF-β1 is often upregulated, contributing to immune evasion by excluding T cells and promoting regulatory T cell differentiation; it is also linked to radioresistance. Consequently, anti-TGF-β1 antibodies are being investigated as therapeutic candidates to enhance the efficacy of radiation therapy and immunotherapies in cancers such as melanoma and breast cancer. Additionally, research in respiratory medicine suggests that TGF-β1 signaling is involved in the dysregulation of bronchial epithelial differentiation in chronic obstructive pulmonary disease (COPD), where it inhibits the generation of ciliated cells and promotes squamous metaplasia.
