Anti-TGF-beta neutralizing antibodies are a class of monoclonal antibodies designed to bind to and inhibit the biological activity of Transforming Growth Factor-beta (TGF-beta) isoforms, primarily TGF-β1, TGF-β2, and TGF-β3. TGF-beta is a pleiotropic cytokine that plays a complex role in disease; while it can inhibit the proliferation of certain cell types, such as B cells in chronic lymphocytic leukemia (CLL), it also acts as a potent immunosuppressor in the tumor microenvironment and promotes epithelial-mesenchymal transition (EMT) and fibrosis. Neutralizing antibodies are used to block these pathological effects, potentially restoring anti-tumor immune responses and inhibiting disease progression. In research settings, these antibodies have been used to demonstrate that TGF-beta acts as an autocrine growth inhibitor in CLL, contributing to the slow progression of the malignant process. This platform is being investigated for various clinical indications, including oncology and fibrotic disorders.
