Anti-TIGITxCTLA-4 BsAb is an experimental bispecific monoclonal antibody that simultaneously targets the inhibitory receptors T‑cell immunoreceptor with Ig and ITIM domains (TIGIT) and cytotoxic T‑lymphocyte–associated protein 4 (CTLA‑4) to enhance antitumor immunity in solid tumors.[6][1] By co-engaging TIGIT and CTLA‑4, the antibody is designed to relieve dual checkpoint–mediated suppression of T cells, promote activation and proliferation of effector T cells, and selectively modulate regulatory T cells within the tumor microenvironment while reducing peripheral immune-related toxicities compared with systemic dual checkpoint blockade.[6][1] Preclinical data indicate that this first‑in‑class CTLA‑4×TIGIT BsAb can preferentially target tumor-infiltrating lymphocytes expressing both checkpoints and improve tolerability in GLP toxicity studies, supporting further development as an immuno-oncology therapy, though a specific corporate sponsor and clinical program have not been clearly disclosed in public sources.[6]
