Anti-TIM-4 is a monoclonal antibody that targets TIM-4 (T-cell immunoglobulin and mucin domain-containing protein 4), an immunoregulatory molecule predominantly expressed on antigen-presenting cells such as macrophages and dendritic cells. The antibody blocks TIM-4's functions, theoretically altering phagocytosis, antigen presentation, and downstream immune responses. In preclinical studies, anti-TIM-4 has been shown to modulate autoimmune disease models (e.g., experimental autoimmune encephalomyelitis, collagen-induced arthritis) and has been considered as a target for cancer immunotherapy to enhance anti-tumor immune responses by affecting the tumor microenvironment and improving efficacy of cancer vaccines[1][2][4]. Its mechanism involves blocking TIM-4-mediated immune tolerance and inhibitory signaling, which can suppress pro-inflammatory cytokine production and alter macrophage activation[1]. As of now, no approved anti-TIM-4 inhibitors or clinical-stage drugs are known, and development remains at the preclinical or mechanistic exploration stage[1][2][4].
