Overview

Anti-Tim-4 antibody refers to a class of experimental monoclonal antibodies that target T-cell immunoglobulin and mucin domain-containing protein 4 (TIM-4), a transmembrane glycoprotein and phosphatidylserine receptor. TIM-4 is primarily expressed on antigen-presenting cells, such as tissue-resident macrophages and dendritic cells, where it plays a critical role in the engulfment of apoptotic cells (efferocytosis) and the regulation of T-cell proliferation. In oncology research, TIM-4+ macrophages have been identified as mediators of immune suppression within the tumor microenvironment. Blockade of TIM-4 using these antibodies has demonstrated the ability to enhance anti-tumor T-cell immunity and synergize with PD-1/PD-L1 checkpoint inhibitors in preclinical models of melanoma and colon cancer. While several research clones like RMT4-53 exist, the class remains in the preclinical stage of development.
