SAGEbio's lead program is an anti-TL1A synbody, a modular biologic engineered as a single-domain synthetic antibody. Developed using AI-guided sequence design and an in-house single-domain antibody library, this therapeutic is designed to selectively block the interaction between Tumor Necrosis Factor-like Ligand 1A (TL1A) and its receptor, Death Receptor 3 (DR3). TL1A is a key mediator and amplifier of chronic inflammation and fibrosis. By inhibiting the TL1A:DR3 axis, the synbody aims to treat inflammatory bowel disease (IBD) and fibrotic diseases while potentially preserving broader immune function. The drug is currently in preclinical development.
