An anti-TLR4 antibody is a monoclonal antibody or antibody fragment designed to target and inhibit Toll-like receptor 4 (TLR4). TLR4 is a pattern recognition receptor that plays a critical role in the innate immune system by detecting pathogen-associated molecular patterns (PAMPs) such as lipopolysaccharide (LPS) from Gram-negative bacteria, as well as damage-associated molecular patterns (DAMPs) released during tissue injury. By binding to TLR4, these antibodies block downstream signaling pathways, including the NF-κB, MAPK, and IRF-3 pathways, thereby suppressing the production of pro-inflammatory cytokines. Anti-TLR4 antibodies, such as paridiprubart (NI-0101), have been investigated for the treatment of various inflammatory and autoimmune conditions, including rheumatoid arthritis, acute kidney injury, and acute respiratory distress syndrome (ARDS).
