Overview

An anti-TLR9 blocking antibody is a therapeutic agent designed to inhibit the activity of Toll-like receptor 9 (TLR9), a key component of the innate immune system that recognizes unmethylated CpG DNA motifs. In the context of oncology, specifically multiple myeloma, tumor cells release mitochondrial DNA (mtDNA) into the bone marrow microenvironment. This mtDNA acts as a damage-associated molecular pattern (DAMP) that triggers TLR9 on immune cells, such as macrophages, inducing the secretion of pro-inflammatory cytokines like IL-6. This inflammatory milieu supports the expansion and progression of the tumor. By utilizing a blocking antibody to antagonize TLR9, researchers aim to disrupt this pro-tumoral signaling pathway, thereby reducing the inflammatory signature of the bone marrow and inhibiting tumor growth.
