The anti-TLR9 intrabody (αT9ib) is an experimental biologic consisting of a single-chain variable fragment (scFv) derived from the monoclonal antibody clone 5G5. It is designed to be expressed intracellularly and localized to the endoplasmic reticulum (ER). Its mechanism involves binding to Toll-like receptor 9 (TLR9) within the ER, thereby preventing the receptor's translocation to endolysosomal compartments where it would normally encounter its ligands (unmethylated CpG DNA). By sequestering TLR9, the intrabody inhibits downstream signaling pathways, including NF-κB activation and the production of pro-inflammatory cytokines like TNFα. This approach is being investigated for the treatment of autoimmune diseases and pancreatic ductal adenocarcinoma (PDAC), where TLR9 signaling contributes to chronic inflammation and tumor progression.
