Anti-TM4SF5 monoclonal antibody is an experimental therapeutic monoclonal antibody targeting transmembrane 4 superfamily member 5 protein (TM4SF5), which is aberrantly expressed in several cancers including hepatocellular carcinoma, colon cancer, and pancreatic cancer. The antibody binds specifically to the extracellular loop 2 (EC2) of TM4SF5, inhibiting its function and suppressing tumor growth, likely by restoring contact inhibition and modulating EMT-related proteins such as E-cadherin and β-catenin. The antibody has shown efficacy in multiple preclinical models, including mouse xenografts for colon and liver cancer, and demonstrates enhanced anti-tumor activity when humanized and combined with agents like sorafenib or doxorubicin. Developers have produced murine (Ab27), chimeric, and humanized (Ab27-hz9) formats, aiming to move toward clinical development as a novel targeted therapy for TM4SF5-expressing cancers[1][2][3][4][5][7].
