Drug intelligence / Profile preview
anti-tMUC1 CAR T cells (also known as MUC28z CAR T cells) are second-generation chimeric antigen receptor (CAR) T cells engineered to target tumor-associated Mucin-1 (tMUC1), an aberrantly glycosylated form of MUC1 selectively expressed on various solid tumors. Developed based on foundational research from the Mukherjee lab at the University of North Carolina at Charlotte and associated with the spinout OncoTab, the CAR construct incorporates a single-chain variable fragment (scFv) derived from the tMUC1-specific monoclonal antibody TAB004, coupled with CD28 costimulatory and CD3ζ signaling domains. In preclinical studies, these CAR T cells have demonstrated potent, antigen-specific cytotoxicity and cytokine release (such as interferon-gamma and granzyme B) against triple-negative breast cancer (TNBC) and pancreatic ductal adenocarcinoma (PDAC) models, while sparing normal tissues expressing healthy MUC1.
