Overview

An **anti-TNF-alpha siRNA** is a small interfering RNA (siRNA) molecule specifically designed to silence the expression of the tumor necrosis factor alpha (TNF-α) gene via the RNA interference (RNAi) pathway. By binding to and promoting the degradation of TNF-α mRNA, it reduces the production of TNF-α protein, a key pro-inflammatory cytokine involved in various inflammatory and autoimmune diseases such as rheumatoid arthritis, inflammatory bowel disease, and acute lung injury. Therapeutic anti-TNF-alpha siRNA is typically delivered using specialized carriers—such as nanoparticles, milk-derived exosomes, or lipid nanoparticles—to protect the siRNA from degradation, facilitate cellular uptake, and permit targeted gene silencing in affected tissues[1][2][3][6][7]. The approach is under preclinical and early clinical investigation, with no product yet approved for clinical use.
