anti-TNF-alpha siRNA (Altamira Therapeutics) is an investigational RNA interference (RNAi) therapy designed to treat a broad range of inflammatory conditions by silencing the gene for tumor necrosis factor-alpha (TNF-α). Developed by Altamira Therapeutics, the drug consists of a small interfering RNA (siRNA) that targets and promotes the degradation of TNF-α messenger RNA (mRNA) after transcription. By specifically preventing the synthesis of the TNF-α protein, a key proinflammatory cytokine, the therapy aims to mitigate the inflammatory cascade at its source. To ensure effective delivery, the siRNA is formulated using Altamira's proprietary OligoPhore platform, which utilizes peptide-based nanoparticles to protect the siRNA from degradation and facilitate its uptake into target cells such as macrophages and synoviocytes. This therapeutic approach is being explored for numerous inflammatory diseases, including rheumatoid arthritis, inflammatory bowel disease, and acute lung injury, with the potential for multiple routes of administration including intra-articular, intravenous, and intranasal.
