Drug intelligence / Profile preview
Anti-TNFR2 antagonist antibodies represent a class of targeted immunotherapies designed to block tumor necrosis factor receptor 2 (TNFR2), a receptor highly expressed on immunosuppressive regulatory T cells (Tregs) and certain tumor cells within the tumor microenvironment (TME). By antagonizing TNFR2, these antibodies inhibit Treg proliferation and survival, thereby reversing immune suppression and promoting the activation and expansion of effector T cells (Teffs). Additionally, they can directly induce cytotoxicity in TNFR2-expressing cancer cells. Mechanistically, some of these antibodies function by stabilizing an inactive, anti-parallel dimeric conformation of TNFR2 on the cell surface, preventing ligand-induced activation of the NF-κB pathway. Several candidates, such as Boston Immune Technologies and Therapeutics' BIR2101 and Apexigen's APX601, have entered clinical and preclinical development for various oncology indications.
