Overview

anti-TP siRNA is a therapeutic modality designed to silence the expression of thymidine phosphorylase (TP), also known as platelet-derived endothelial cell growth factor (PD-ECGF). TP is an enzyme that plays a critical role in pyrimidine nucleoside metabolism and is frequently overexpressed in various solid tumors, where it promotes angiogenesis, inhibits apoptosis, and contributes to tumor progression and metastasis. By utilizing the RNA interference (RNAi) pathway, anti-TP siRNA molecules specifically bind to and induce the degradation of TP mRNA, thereby reducing TP protein levels. This inhibition aims to suppress tumor-associated angiogenesis and sensitize cancer cells to certain chemotherapeutic agents, such as fluoropyrimidines. While primarily investigated in preclinical oncology models, various delivery systems, including nanoparticles and viral vectors, are being explored to enhance the stability and uptake of these siRNA molecules in vivo.
