anti-TRBC1 CAR-T is a chimeric antigen receptor (CAR) T-cell therapy designed to specifically target and kill T-cell malignancies that express the T cell receptor β-chain constant region 1 (TRBC1). This therapy involves genetically engineering T cells to express a CAR that recognizes TRBC1. A key challenge identified is that over-expressed CARs on anti-TRBC1 CAR-transduced TRBC1+ T cells (CAR-C1) can bind to autologous TRBC1, masking the target from other anti-TRBC1 CAR-T cells and reducing therapeutic potency. Additionally, co-culture with TRBC1+ cells can lead to CAR-T cell exhaustion and terminal differentiation. To overcome these issues, pre-depletion of TRBC1+ T cells before CAR-T manufacturing is proposed to enhance efficacy and prevent resistance and exhaustion.
