Anti-TRBC1 CAR-T cell therapy is an investigational chimeric antigen receptor T-cell (CAR-T) therapy developed by Shanghai General Hospital, affiliated with the Shanghai Jiao Tong University School of Medicine. This cell-based immunotherapy specifically targets the T-cell receptor beta-chain constant region 1 (TRBC1). In humans, mature T cells express either TRBC1 or TRBC2 in a mutually exclusive manner. Because T-cell malignancies are clonal, they typically express only one of these constant regions. By targeting TRBC1, the therapy is designed to selectively eliminate malignant T cells while sparing the healthy TRBC2-positive T-cell population, thereby potentially preserving a portion of the patient's cellular immune function. It is currently being evaluated in Phase 1 clinical trials for various relapsed or refractory TRBC1-positive T-cell hematological malignancies, including peripheral T-cell lymphoma, angioimmunoblastic T-cell lymphoma, and T-lymphoblastic lymphoma.
