Overview

Anti-TRBC1 CAR-T cells (Zhejiang University) is an investigational chimeric antigen receptor (CAR) T-cell therapy being developed by the Second Affiliated Hospital, School of Medicine, Zhejiang University for the treatment of relapsed or refractory peripheral T-cell lymphoma (PTCL). The therapy utilizes autologous T-cells engineered to target the T-cell receptor beta-chain constant domain 1 (TRBC1) or 2 (TRBC2). In humans, the T-cell receptor beta chain constant region is encoded by two nearly identical genes, TRBC1 and TRBC2, which are expressed in a mutually exclusive manner on the surface of T-cells. Since T-cell lymphomas are monoclonal, the malignant cells express only one of these domains. By targeting the specific TRBC isoform expressed by the lymphoma, this CAR-T approach selectively eradicates malignant cells while sparing the healthy T-cell population that expresses the alternative TRBC isoform, thereby preserving a portion of the patient's adaptive immunity and reducing the risk of pan-T-cell aplasia. It is currently being evaluated in Phase I clinical trials.
