anti-TRBC1-MMAE is an experimental antibody-drug conjugate (ADC) comprising a monoclonal antibody against TRBC1, a constant region of the T-cell receptor β-chain expressed clonally in certain T cell leukemias and lymphomas, conjugated to the cytotoxic payload monomethyl auristatin E (MMAE) via a cleavable linker. The anti-TRBC1 antibody specifically targets TRBC1+ cancer cells, allowing internalization and selective delivery of MMAE, which is released intracellularly to disrupt microtubule polymerization, leading to apoptosis of the targeted tumor cell. The drug was developed in a research setting primarily by a team at Johns Hopkins, for the treatment of TRBC1+ T cell malignancies, demonstrating potent and selective killing of TRBC1+ T-cell leukemias and lymphomas in preclinical models[2][3][6].
