anti-Trop2 ETA is a recombinant immunotoxin (rIT) designed for the targeted treatment of Trop2-overexpressing epithelial tumors, including cervical, breast, and gastrointestinal cancers. Developed by researchers at the University of Cape Town, the molecule consists of a single-chain variable fragment (scFv) specific for Tumor-associated calcium signal transducer 2 (Trop2) fused to the N-terminus of Pseudomonas exotoxin A (ETA). Upon binding to Trop2 on the cell surface, the immunotoxin is internalized, where the ETA payload inhibits protein synthesis by ADP-ribosylating elongation factor 2, leading to cell death. Preclinical studies have demonstrated antigen-specific, dose-dependent killing of Trop2-positive tumor cell lines in vitro, with ongoing evaluations focused on in vivo pharmacokinetics and biological activity.
