anti-TROP2 HCAb is a novel heavy chain-only antibody (HCAb) targeting Trophoblast cell surface antigen-2 (TROP2), developed by KisoJi Biotechnology for the treatment of epithelial cancers. Isolated using a proprietary transgenic HCAb mouse platform, these antibodies exhibit picomolar binding affinity to human TROP2 and demonstrate potent anti-tumor activity as naked antibodies. Unlike traditional TROP2-targeted antibody-drug conjugates (ADCs), these HCAbs directly inhibit tumor cell viability and induce tumor regression in vivo without the need for a toxin payload. Furthermore, glycoengineering to an afucosylated form can enhance their antibody-dependent cellular cytotoxicity (ADCC) activity. The antibodies target non-overlapping epitopes on TROP2 and show high specificity for human and cynomolgus TROP2 over other species.
