Anti-Trop2 U-CAR-NK cells are an investigative allogeneic (universal) cell therapy consisting of natural killer (NK) cells engineered to express a chimeric antigen receptor (CAR) targeting Trophoblast cell-surface antigen 2 (Trop2). Trop2, also known as Tumor-associated calcium signal transducer 2 (TACSTD2), is a transmembrane glycoprotein overexpressed in various epithelial malignancies, including non-small cell lung cancer (NSCLC), where it facilitates tumor growth and metastasis. The "universal" (U) designation signifies that these cells are derived from healthy donors and are prepared as an "off-the-shelf" product, potentially undergoing gene editing to reduce the risk of graft-versus-host disease (GvHD) and enhance persistence. Upon administration, these CAR-NK cells are designed to recognize and bind to Trop2-expressing tumor cells, leading to direct cytotoxic cell killing through the release of perforins and granzymes.
