Anti-TSPAN5 antibody refers to monoclonal antibodies targeting Tetraspanin-5 (TSPAN5), a cell-surface protein belonging to the TspanC8 subfamily. TSPAN5 plays a critical role in regulating the trafficking, maturation, and cell-surface expression of the metalloprotease ADAM10, which in turn activates Notch signaling. In the tumor microenvironment, TSPAN5 is highly expressed by tumor-associated macrophages (TAMs) and promotes mammary tumor growth, invasion, and metastasis. Preclinical research, primarily led by the University of Sheffield and Inserm, has demonstrated that antibody-mediated ablation or knockdown of TSPAN5 can inhibit primary tumor growth, promote a pro-inflammatory macrophage phenotype, and enhance survival in breast cancer models. Additionally, anti-TSPAN5 antibodies have shown potential in inhibiting ligand-induced Notch signaling in other Notch-driven cancers.
