Overview
anti-TWEAK refers to a class of monoclonal antibodies designed to neutralize Tumor Necrosis Factor-like Weak Inducer of Apoptosis (TWEAK), a cytokine also known as TNFSF12. TWEAK signals through its cognate receptor, Fn14 (Fibroblast growth factor-inducible 14), to promote inflammation, angiogenesis, and tissue remodeling. In autoimmune conditions like psoriasis, TWEAK acts synergistically with IL-17 and TNF to drive the expression of inflammatory chemokines and cytokines in keratinocytes. Biogen has been a primary developer of anti-TWEAK therapeutics, notably BIIB023, which was evaluated in clinical trials for lupus nephritis and rheumatoid arthritis. Although clinical development for those indications was discontinued, research continues into the role of TWEAK inhibition in other inflammatory and fibrotic diseases.
