Anti-Unc5B antibody refers to a class of research-stage monoclonal antibodies designed to target and inhibit the Unc5B receptor (also known as UNC5H2 or p53RDL1), a netrin-1 receptor that functions as a dependence receptor. Unc5B plays a critical role in regulating vascular branching during angiogenesis and maintaining the integrity of the blood-brain barrier (BBB). By acting as an antagonist, these antibodies block Unc5B signaling, which has been shown to induce transient, size-selective BBB permeability. This mechanism is being explored to enhance the delivery of chemotherapeutic agents (such as doxorubicin and temozolomide) and targeted therapies (such as osimertinib) to the central nervous system, particularly in the treatment of glioblastoma. Additionally, anti-Unc5B antibodies are being investigated for their potential to modulate pathological angiogenesis in conditions such as solid tumors, atherosclerosis, age-related macular degeneration (AMD), and inflammatory diseases like rheumatoid arthritis and psoriasis.
