Overview

anti-uPAR CAR T is an experimental chimeric antigen receptor (CAR) T-cell therapy targeting the urokinase plasminogen activator receptor (uPAR). uPAR is a cell surface protein that is significantly upregulated in recurrent glioblastoma (GBM), particularly on brain tumor initiating cells (BTICs) which are associated with tumor recurrence and resistance to standard therapies. Developed by McMaster University and the National Research Council of Canada, this CAR T-cell therapy utilizes novel anti-uPAR single-domain antibodies (sdAbs) to direct T-cell cytotoxicity against uPAR-expressing tumor cells. Preclinical data presented at AACR 2024 demonstrated that these CAR T cells exhibit potent anti-tumor activity in vitro and significantly reduce tumor burden while extending survival in patient-derived xenograft models of GBM.
