Overview

Anti-uPAR CAR-T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target the urokinase plasminogen activator receptor (uPAR), also known as CD87. uPAR is a cell-surface protein that is significantly upregulated on senescent cells and in various cancers, including glioblastoma, ovarian cancer, and brain metastases. The therapy involves engineering T cells to express a CAR that specifically binds to uPAR, leading to the targeted destruction of cells expressing this receptor. Developed by researchers at Memorial Sloan Kettering Cancer Center and McMaster University, this modality is being explored for two primary therapeutic purposes: as a senolytic to treat age-related conditions (such as metabolic dysfunction and fibrosis) and as an oncology treatment to target tumor cells and the immunosuppressive tumor microenvironment. Preclinical studies in mouse models have shown that anti-uPAR CAR-T cells can effectively reduce tumor burden and extend survival.
