Anti-VCAM-1 antibodies are a class of monoclonal antibodies that target Vascular Cell Adhesion Molecule-1 (VCAM-1, also known as CD106), a cell surface protein primarily expressed on inflamed vascular endothelium. VCAM-1 plays a pivotal role in the recruitment of leukocytes (such as monocytes and lymphocytes) from the blood into tissues by interacting with the integrin alpha-4 beta-1 (VLA-4) expressed on the leukocyte surface. By blocking this interaction, anti-VCAM-1 antibodies aim to reduce vascular inflammation and are being investigated for the treatment of conditions such as atherosclerosis, asthma, rheumatoid arthritis, and inflammatory bowel disease. In oncology, research has demonstrated that VCAM-1 blockade can disrupt the survival signals provided by tumor-associated myeloid cells to T-cell acute lymphoblastic leukemia (T-ALL) blasts. While several candidates, including Hanwha Chemical's HD101, have been studied in preclinical models, no anti-VCAM-1 antibody has yet reached the market.
