Overview

anti-Vcan siRNA nanoparticles are a preclinical therapeutic candidate designed to target the bone marrow microenvironment in multiple myeloma. The drug consists of small interfering RNA (siRNA) molecules encapsulated in nanoparticles, specifically targeting the expression of Versican (Vcan), an extracellular matrix proteoglycan. In preclinical models, Vcan is highly expressed by pro-tumoral myeloma-associated macrophages (MAMs). By silencing Vcan, these nanoparticles modulate the immune microenvironment, leading to a reduction in the expression of immune checkpoint molecules such as PD-L1 on macrophages and PD-1 on CD4+ and CD8+ T cells, thereby potentially overcoming immunosuppression and enhancing anti-tumor immunity.
