anti-VEGFR2 + anti-CD3ε is a bispecific T-cell engaging antibody (bsAb) designed to target the tumor vasculature and overcome endothelial cell anergy. It simultaneously binds to Vascular Endothelial Growth Factor Receptor 2 (VEGFR2), which is highly expressed on tumor endothelial cells, and the CD3ε subunit of the T-cell receptor complex on effector T cells. By cross-linking T cells with the tumor endothelium, the antibody triggers T-cell activation and the localized release of proinflammatory cytokines such as TNF-α, IFN-γ, and IL-6. This inflammatory response upregulates adhesion molecules (including E-selectin, ICAM-1, and VCAM-1) on otherwise quiescent tumor endothelial cells, thereby enhancing T-cell transendothelial migration and infiltration into the tumor microenvironment. This mechanism is intended to improve the efficacy of cancer immunotherapy by facilitating the entry of effector cells into the tumor parenchyma.
