Anti-VEGFR2 chimeric antigen receptor CD8+ peripheral blood lymphocytes is an investigational adoptive cell therapy consisting of autologous CD8+ T cells genetically modified to express a chimeric antigen receptor (CAR) targeting vascular endothelial growth factor receptor 2 (VEGFR2). Unlike most CAR-T cell therapies that target antigens expressed directly on tumor cells, this agent is designed to target the tumor vasculature. VEGFR2 is a primary mediator of angiogenesis and is highly overexpressed on the endothelial cells of the tumor blood vessels compared to normal tissue. By targeting VEGFR2, these engineered lymphocytes are intended to recognize and eliminate tumor-associated endothelial cells, thereby disrupting the blood supply to the tumor and inducing tumor necrosis and regression. This therapy was primarily developed and evaluated in clinical trials by the National Cancer Institute (NCI) for the treatment of various metastatic solid tumors.
