anti-VISTA refers to monoclonal antibodies or therapeutics that target VISTA (V-domain immunoglobulin suppressor of T cell activation, also known as PD-1H, C10orf54, VSIR, B7-H5, DD1α, Gi24, Dies1), a negative immune checkpoint regulator highly expressed on myeloid cells (such as monocytes, dendritic cells) and to a lesser extent on T lymphocytes[4][5][6][7]. anti-VISTA drugs block the VISTA-mediated signaling pathway, which suppresses T cell activation and promotes immune evasion by tumors. By inhibiting VISTA, these drugs aim to reverse immune suppression, restore T cell function, and enhance antitumor immunity, making them potential cancer immunotherapies. Mechanistically, the antibody backbone and isotype influence which immune cells are stimulated, and therapeutic efficacy often involves specific Fc-receptor interactions as well as binding under acidic conditions typical of the tumor microenvironment[1][6]. Notable examples of anti-VISTA drugs in clinical development include HMBD-002, KVA12123, CI-8993 (JNJ-61610588), and SNS-101[5][6][7]. Primary indications are cancer types such as melanoma, pancreatic cancer, NSCLC, prostate, ovarian, squamous cell carcinoma, colorectal cancer, and more[4][5].
