anti-VISTA-IgG1 is an experimental monoclonal antibody targeting V-domain Ig suppressor of T cell activation (VISTA), also known as PD-1H. Developed by Boehringer Ingelheim, this antibody is engineered with a human IgG1 isotype to leverage Fc-gamma-receptor (FcγR) interactions for immune stimulation. Unlike its IgG4 counterpart, the IgG1 variant is capable of activating resting immune cells, such as upregulating HLA-DR on myeloid cells, in addition to augmenting ongoing immune responses. Its activity is strictly dependent on Fc-receptor binding, suggesting a mechanism that involves cross-linking or effector cell engagement to overcome VISTA-mediated immunosuppression in the tumor microenvironment. Research has explored its potential in treating hematological malignancies like acute myeloid leukemia (AML) and other oncological indications.
