anti-VISTA-IgG4

anti-VISTA-IgG4 is a monoclonal antibody targeting V-domain Ig suppressor of T cell activation (VISTA), also known as PD-1H. VISTA is an immune superfamily cell surface molecule primarily expressed on myeloid cells, but also on NK cells and T cells, where it functions as an immune checkpoint by providing inhibitory signals. Research conducted by Boehringer Ingelheim has demonstrated that the IgG4 isotype of anti-VISTA antibodies can effectively augment ongoing immune reactions, such as cytokine production (e.g., TNF), in a manner dependent on Fc-gamma-receptor (FcγR) interaction. Unlike IgG1 variants, which may non-specifically activate resting immune cells, the IgG4 isotype appears to specifically enhance existing immune responses. This profile suggests potential therapeutic utility in oncology, particularly in conditions like acute myeloid leukemia (AML), by promoting immune cell interactions with cancer cells.
