The **anti-VISTA x MSLN bispecific antibody** is an experimental immunotherapy developed by **Y-Biologics** for the treatment of **epithelioid malignant pleural mesothelioma (MPM)**. This bispecific antibody (BsAb) simultaneously targets **Mesothelin (MSLN)**, a tumor-associated antigen highly expressed in epithelioid MPM, and **V-domain Ig suppressor of T cell activation (VISTA)**, an immune checkpoint protein that contributes to the immunosuppressive tumor microenvironment. By dual-targeting, the antibody aims to localize to the tumor site via MSLN binding while blocking VISTA-mediated immune evasion. Preclinical studies indicate that the BsAb exhibits improved binding affinity and antibody-dependent cell cytotoxicity (ADCC) activity compared to monotherapy. It also serves as the structural foundation for a trispecific immunocytokine, created by fusing the BsAb with an interferon-beta (IFN-β) mutein to further enhance anti-tumor immune responses through both direct cytotoxicity and indirect activation of immune cells.
