The anti-VISTA x MSLN bispecific antibody-interferon beta mutein fusion protein is a trispecific immunocytokine developed for the treatment of epithelioid malignant pleural mesothelioma (MPM). It consists of a bispecific antibody targeting the immune checkpoint V-domain Ig suppressor of T cell activation (VISTA) and the tumor differentiation antigen Mesothelin (MSLN), fused to a stabilized interferon-beta (IFN-β) mutein containing R27T and C17S mutations. This fusion protein is designed to leverage the co-expression of VISTA and MSLN in epithelioid MPM to localize IFN-β activity to the tumor microenvironment, thereby enhancing anti-tumor efficacy while minimizing systemic toxicity. The drug acts through multiple mechanisms, including direct cytotoxicity from IFN-β, antibody-dependent cell cytotoxicity (ADCC) mediated by the antibody component, and the activation of immune cells via VISTA blockade and IFN-β signaling.
