The anti-whitlow monoclonal antibody is a novel biologic tool developed to target the Whitlow linker peptide, a 15-amino acid sequence (typically (Gly4Ser)3) frequently incorporated into the single-chain variable fragments (scFvs) of chimeric antigen receptor (CAR) constructs. Developed by a collaborative team at the Fred Hutchinson Cancer Center and the University of Washington, these antibodies serve as a universal detection reagent for CAR T-cells, independent of their specific antigen targets (such as CD19, BCMA, or CD33). The antibody is applicable in flow cytometry for monitoring CAR T-cell persistence in blood, immunohistochemistry for spatial characterization in tissues, and manufacturing processes for the selective enrichment of CAR-expressing cells. Its ability to bind the linker framework makes it a versatile asset for both preclinical research and clinical translational studies.
