Overview

The **anti-Wnt-2 monoclonal antibody** is an investigational biologic that specifically binds and blocks the activity of the Wnt-2 protein, a secreted ligand involved in the canonical Wnt/β-catenin signaling pathway. Dysregulation of this pathway is implicated in oncogenesis, particularly in tumors such as melanoma, non-small cell lung cancer (NSCLC), colorectal cancer (CRC), and esophageal squamous cell carcinoma (OSCC). The antibody induces apoptosis in Wnt-2–overexpressing cancer cells by downregulating survival pathways, reducing levels of anti-apoptotic proteins (e.g., survivin), and inhibiting β-catenin signaling. It also reverses immune suppression within the tumor microenvironment by blocking CAF-derived WNT2, enhancing dendritic cell differentiation and CD8+ T-cell–mediated antitumor immunity, especially when combined with checkpoint blockade (anti–PD-1). Studies indicate efficacy in preclinical models for both direct tumor cytotoxicity and immunomodulation, and a favorable tumor-selective binding profile has been reported[2][4][5][8][9].
