Drug intelligence / Profile preview
Anti-ZIKV nanobodies are single-domain antibodies (VHH) developed for the prevention and treatment of Zika virus (ZIKV) infection [5740]. Developed by researchers at the University of Hong Kong, these nanobodies were identified from a library constructed by immunizing alpacas with recombinant ZIKV proteins [5740]. They target either the ZIKV envelope (E) protein or nonstructural protein 1 (NS1), demonstrating potent neutralizing and antiviral activity in vitro and in vivo [5740]. In preclinical mouse models, treatment with these nanobodies significantly improved survival rates and reduced viral burden in brain tissues when administered as prophylaxis or therapy [5740]. Crucially, unlike conventional monoclonal antibodies, these nanobodies do not induce antibody-dependent enhancement (ADE) of infection and have been shown to downregulate interleukin-1-beta (IL-1beta) levels [5740].
