**antibody 211-14** is a **human neutralizing monoclonal antibody** specifically targeting secreted-form ADAM28 (a disintegrin and metalloproteinase 28), which is overexpressed in many non–small cell lung carcinomas (NSCLC)[1][3][9]. ADAM28 promotes tumor proliferation and metastasis through reactivation of insulin-like growth factor-1 (IGF-1) and resistance to von Willebrand factor (VWF)–induced apoptosis by digesting IGF-binding protein-3 and VWF. antibody 211-14 binds to the junctional region between the cysteine-rich domain and the secreted-specific domain of ADAM28s and acts as a neutralizing inhibitor. This antibody suppresses IGF-1–dependent cancer cell proliferation, enhances cancer cell susceptibility to VWF-induced apoptosis, inhibits lung tumor growth, and reduces metastasis in preclinical models. It demonstrated strong efficacy as both monotherapy and in combination with docetaxel for NSCLC in animal studies. The antibody is highly specific for human and monkey ADAM28s but does not recognize membrane-anchored mouse ADAM28m. No adverse effects were observed at doses up to ten times higher than the effective dose in mice[1][3][9].
