Drug intelligence / Profile preview
AntiFib-rtPA6 is an experimental, preclinical-stage fibrin-targeted fusion protein designed for the treatment of arterial and venous thrombosis, including pulmonary embolism (PE) and acute ischemic stroke. Developed by researchers at INSERM and the Etablissement Français du Sang (EFS), the molecule consists of a high-affinity, fibrin-specific monoclonal antibody fused to two catalytic domains of recombinant tissue plasminogen activator (rtPA) via non-cleavable linkers. This targeted approach concentrates thrombolytic enzymatic activity directly within fibrin-rich thrombi, enhancing local efficacy while minimizing systemic exposure to reduce the risk of bleeding and neurotoxicity. In ex vivo studies, AntiFib-rtPA6 demonstrated strong and specific binding to human and murine fibrin without targeting fibrinogen, and achieved thrombus lysis in patient-derived PE and stroke samples comparable to free rtPA.
