Overview

This is a personalized cell therapy in which cytotoxic T lymphocytes (CTLs) are generated ex vivo by stimulating patient-derived dendritic cells (DCs) that have been infected or transduced with a recombinant adeno-associated virus (rAAV) vector encoding the carcinoembryonic antigen (CEA) gene. The process involves isolating DCs from the patient, infecting them with rAAV carrying the CEA gene so they express and present CEA peptides on their surface, and then co-culturing these modified DCs with autologous T cells to induce expansion of antigen-specific CTLs targeting CEA-expressing tumor cells. These CTLs are then infused back into the patient as an adoptive immunotherapy. The mechanism of action relies on enhanced presentation of tumor-associated antigens (specifically CEA), leading to activation and proliferation of CD8+ cytotoxic T lymphocytes capable of recognizing and killing cancer cells expressing CEA. This approach is being explored primarily for cancers that overexpress carcinoembryonic antigen, such as colorectal cancer and other epithelial tumors[1][6][7][9].
