Drug intelligence / Profile preview
Antisense oligonucleotides targeting circPHAF1 are synthetic oligonucleotides designed to inhibit the circular RNA circPHAF1. Developed by researchers at The First Affiliated Hospital of Guangzhou Medical University, these antisense oligonucleotides (ASOs) are being investigated for the treatment and prevention of calcium oxalate urolithiasis (kidney stones). Mechanistically, circPHAF1 is upregulated in kidney tissues during calcium oxalate stone formation, where it binds to general transcription factor IIF subunit 1 (GTF2F1) and recruits it to the C-C motif chemokine ligand 2 (CCL2) promoter, leading to sustained CCL2 activation and macrophage recruitment. Additionally, circPHAF1 is packaged into extracellular vesicles (EVs) and internalized by macrophages, promoting their polarization toward the inflammatory M1 phenotype. By targeting and degrading circPHAF1, these ASOs exert potent inhibitory effects on renal calcium oxalate crystal formation and associated inflammatory injury in vivo.
