Drug intelligence / Profile preview
Antisense oligonucleotides targeting GRB7-S are synthetic oligonucleotide therapeutics designed to target and degrade the mRNA of the short isoform of Growth Factor Receptor-Bound Protein 7 (GRB7-S, characterized by exon 13 skipping) [EP100]. GRB7-S is highly expressed in early-onset gastric cancer (EOGC) tissues and is associated with poor prognosis, promoting epithelial-mesenchymal transition (EMT), cell invasion, and distant metastasis [EP100]. Mechanistically, Musashi RNA-binding protein 1 (MSI1) induces the skipping of exon 13 in GRB7 pre-mRNA, activating the Kras/PI3K/AKT/mTOR signaling pathway [EP100]. By targeting and degrading the GRB7-S transcript, these antisense oligonucleotides suppress MSI1-driven metastasis, offering a potential therapeutic strategy for early-onset gastric cancer [EP100].
