ANV321 (also referred to as ANV322) is a second-generation small molecule inhibitor of DPAGT1 (UDP-N-acetylglucosamine—dolichyl-phosphate N-acetylglucosaminephosphotransferase), developed by the biotech company Anviron in collaboration with researchers at the University of Tennessee Health Science Center. As a capuramycin analogue, ANV321 targets the N-glycan biosynthesis pathway, which is often upregulated in various cancers. The drug is primarily being developed to treat drug-resistant HER2-positive breast cancer and other solid tumors that express DPAGT1. It has shown potential in overcoming resistance to standard therapies like trastuzumab by inhibiting the enzymatic activity of DPAGT1, thereby affecting the glycosylation and stability of key oncogenic receptors.
