Overview

ANV419 is a novel antibody–cytokine fusion protein designed as a selective interleukin-2 (IL-2) agonist. It consists of human IL-2 fused to an anti-IL-2 monoclonal antibody that sterically blocks IL-2 from binding to the α subunit (CD25) of its receptor, while retaining high affinity for the βγ subunits (CD122/CD132). This selectivity enables preferential stimulation and expansion of CD8+ effector T cells and natural killer (NK) cells, which are associated with antitumor activity, while minimizing activation of immunosuppressive regulatory T cells. ANV419 has demonstrated favorable safety and tolerability in early-phase clinical trials, with manageable adverse events and no dose-limiting toxicities up to 243 µg/kg. It is being developed primarily for advanced solid tumors and hematological malignancies as both monotherapy and in combination with other agents such as immune checkpoint inhibitors.
