Overview

ANV419 + anti-PD-1 antibody is an **investigational combination immunotherapy** currently in clinical trials for advanced melanoma and other solid tumors. ANV419 is an **antibody-cytokine fusion protein** consisting of interleukin-2 (IL-2) fused to an anti-IL-2 monoclonal antibody, designed to preferentially stimulate CD8+ T cells and natural killer (NK) cells, while limiting regulatory T cell (Treg) expansion. The combination involves co-administration with an anti-PD-1 antibody (such as pembrolizumab), enhancing antitumor immune responses by both stimulating effector immune cells and blocking immune checkpoint pathways. The combination is being studied for synergistic activity and improved tolerability compared to high-dose IL-2 or PD-1 blockade alone.[5][3][7]
