Overview

ANX-514 is a detergent-free, nano-emulsion reformulation of the taxane chemotherapy drug docetaxel. Developed by Mast Therapeutics (formerly Adventrx Pharmaceuticals), it was designed to eliminate the need for polysorbate 80 (Tween 80), a detergent used in the standard docetaxel formulation (Taxotere) that is frequently associated with severe hypersensitivity reactions and requires corticosteroid premedication. ANX-514 was intended to improve the safety profile and patient convenience of docetaxel therapy by delivering the active ingredient in a novel nano-emulsion. However, the program was discontinued after a pivotal Phase 1 bioequivalence study in patients with advanced cancer failed to meet its primary pharmacokinetic endpoints in 2009, as the formulation did not demonstrate bioequivalence to Taxotere.
