Overview

ANX1502 is a first-in-class, orally administered small molecule prodrug that inhibits the classical complement pathway by selectively targeting the active form of Complement component 1s subcomponent (C1s), an essential enzyme in this cascade. Upon administration, ANX1502 is converted in the body to its active compound, ANX1439. By blocking C1s activity, it prevents cleavage of complement proteins C4 and C2, thereby halting downstream activation events that drive inflammation and tissue damage in complement-mediated autoimmune diseases such as cold agglutinin disease (CAD). Developed by Annexon, ANX1502 has demonstrated good tolerability and pharmacokinetic properties in Phase 1 clinical trials with healthy volunteers. The drug is being advanced into proof-of-concept studies for CAD and may have broader applications across serious autoimmune conditions[1][3][4][5][6][7].
