ANXA1 peptide Ac2-26 is a synthetic peptide mimicking the N-terminal 25 amino acids of annexin A1, a protein regulated by glucocorticoids with anti-inflammatory and pro-resolving effects. Ac2-26 acts primarily by binding and activating formyl peptide receptors FPR1 and FPR2 (also called FPR2/ALX), triggering downstream ERK1/2 phosphorylation and modulating inflammatory cell responses such as migration and cytokine production. In preclinical models, Ac2-26 has demonstrated anti-inflammatory, tissue repair, and immunomodulatory actions across conditions such as inflammatory bowel disease, lung injury, diabetic wounds, and models of COVID-19 cytokine storm. It exhibits chemokinetic effects on leukocytes and promotes resolution of inflammation by modulating neutrophil and macrophage activity, reducing pro-inflammatory cytokines (e.g., TNF-α, IL-1β, IL-6), and enhancing anti-inflammatory signaling (e.g., IL-10, TGF-β).
